Through the study of fermentation conditions on chitin Degradation Strain HD001, determine the most suitable medium of Producing Chitinase were divided into (w/v): (NH4) 2SO4 2%, NaCl 0.5%, K2HPO4 0.07%, KH2P O4 0.03%, MgSO4 7H2O 0.05% and 0.1% glucose, 0.3% yeast extract and colloid chitin 1.5%; the optimum production enzyme culture conditions: medium initial pH value 6, culture temperature is 30 degrees centigrade, the incubation time is 120 h, fermentation enzyme activity reached 376.2 U/mL. by ammonium sulfate precipitation, DEAE- cellulose ion exchange chromatography and Sephacryl S-100 gel filtration chromatography of the enzyme was purified after SDS-PAGE electrophoresis showed a single band with the relative molecular mass about 85.7 kDa.
